The absence of basal transcriptional level of this gene in humans was suggested to be due to the absence of the main core promoter elements; CAAT and
TATA boxes (Leung, Rajendran, Monfries, Hall, and Lim, 1990).
The most characteristic cis-regulatory element, detected in the mGH, is the
TATA box (Goldberg-Hogness box), a binding site of the TATA-Binding Protein (TBP) that is part of the transcription preinitiation complex (Schluesche et al., 2008).
Third, the sequence features, including CpG density, conservation score,
TATA box affinity, and normalized tag counts, were calculated around the putative TSSs regions and the random regions.
The Mad-1 NCCR is composed of two 98-bp tandem repeats, each containing a
TATA box (Figure 1).
For construction of luciferase vector containing only promoter (-2,657 to +39) of porcine [beta]-casein gene, PCR was conducted using porcine [beta]-casein 5' arm DNA containing normal TATA or mutant
TATA box as a template.
Then, the alignment was constrained using the Cis elements (G-Box, H-Box and
TATA Box) as guides.
The sequence also has a
TATA box located between 15 and 25 nucleotides in the 3' direction of the closest Dof element.
